Freshwater calanoid copepods in Southern Luzon identified by graduate students at the University of Santo Tomas from 2011-2015.Sampling Description
Study Extent
The extent of the study conducted is primarily composed of multiple undergraduate and graduate theses conducted from 2011 up to 2015 which involved lakes in Southern Luzon.Sampling
Plankton samples were collected using a conical plankton net of 100 μmd in mesh size during several field sampling trips. Samples were fixed in 95% ethyl alcohol.Method steps
- Samples were allowed to fix for at least 10 days prior to sample processing. Afterwhich, samples were sieved using 70 μm and 45 μm sieves and fixed in 70% ethyl alcohol with Rose Bengal dye. Calanoids were sorted and identified to the lowest possible taxonomic classification. Dissection was done using fine tungsten needles under a BestScope Stereomicroscope. Specimens were mounted in slides using glycerin as a mounting medium and sealed with a cover slip using clear nail polish as sealant. Further taxonomic observations and line drawings were made using an Olympus CX21 compound microscope with an attached drawing tube. Adobe Photoshop CC and Adobe Illustrator were used to produce digital line drawings of the specimens. Finer details were viewed under a Optika B-500 Phase Contrast Microscope and the Optika Software.
